Using Spherical Harmonic Virtual Screening Tools to Compare and Classify HIV Entry Inhibitors for the CXCR4 and CCR5 Co-Receptors

چکیده
HIV entry inhibitors have emerged as a new generation of anti-retroviral drug that block viral fusion with the CXCR4 and CCR5 membrane co-receptors. In the last few years, many such entry inhibitors have been developed, and several are currently in clinical trials. However, because no crystal structures for the co-receptor proteins are available, the binding modes of the known inhibitors within the coreceptor extracellular pockets need to be analyzed using site-directed mutagenesis and computational experiments.

Peptide and protein-based inhibitors of HIV-1 co-receptors.
Human immunodeficiency virus (HIV) afflicts an estimated 30 million people globally, making it a continuing pandemic. Despite major research efforts, the rate of new infections has remained relatively static over time. This article reviews an emerging strategy for the treatment of HIV, the inhibition of the co-receptors necessary for HIV entry, CCR5 and CXCR4. The aim of this article is to high...

Chemokines are small proteins (7-16kDa) that act at G protein-coupled receptors to regulate a variety of physiological and pathophysiological processes1,2). The human immunodeficiency virus Type 1 (HIV-1) principally uses CCR5 and CXCR4 chemokine receptors as coreceptors with CD4 to gain entry into target cells3,4). Therefore, a molecule that binds to the CCR5 receptor could potentially prevent...
متن کاملFrequency of CCR5?32 Variant in North-West of Iran
Chemokine and chemokine receptors show several variations which may affect resistance to infectious disease. A 32 base pair deletion in the open reading frame of the human CCR5 gene (CCR5?32) results in producing a truncated antigen which fails to be presented on the surface of target cells. CCR5?32 variant is not a functional co receptor for HIV-1 entrance and delay the onset of acquired immun...
متن کاملA possible role for CCR5 in the progression of atherosclerosis in HIV-infected patients: a cross-sectional study
BACKGROUND Chemokines can block viral entry by interfering with HIV co-receptors and are recognised mediators of atherosclerosis development. A number of experimental drugs that inhibit HIV entry arrest the development of atherosclerosis in animal models. We hypothesised that the expression of chemokine receptors in circulating leukocytes is associated with the rate of atherosclerosis progressi...
متن کاملThe Puzzling Role of CXCR4 in Human Immunodeficiency Virus Infection
The human immunodeficiency virus type-1 (HIV-1) is the etiological agent of the acquired immunodeficiency syndrome (AIDS), a disease highly lethal in the absence of combination antiretroviral therapy. HIV infects CD4(+) cells of the immune system (T cells, monocyte-macrophages and dendritic cells) via interaction with a universal primary receptor, the CD4 molecule, followed by a mandatory inter...
